To get Lami Staff, you must progress and reach Act 6 of the Water Park Map in Story Mode. Once you get there, you must survive and win all 15 Waves to get the Lami Staff in Anime Last Stand.

It is not confirmed that you will get this resource after completing the wave. However, if you play it on the Nightmare Difficulty, there is a 25% chance that you will be rewarded with Lami Staff. This makes it pretty clear that playing on Normal difficulty has relatively lower chances of giving you the Lami Staff.
