The Dragon-Hunter's Great Katana can be obtained by defeating the Ancient-Dragon Man boss in the Dragon's Pit dungeon, which is located east of the Gravesite Plain Site of Grace (the first Grace you find in the DLC). This boss is located at the end of the dungeon and defeating him will reward you with this Great Katana.
The Ancient Dragon Man is a powerful boss that can be defeated in a few hits with his jumping slash attack and his forward slash attack. Your best bet is to corner him and damage him with some heavy melee combos. He also does Dragon Bite and Magma Breath attacks, which you should stay away from.
You can also use magic spells on him from a longer distance, but he gets close very quickly, so you'll have to dodge and roll quickly. He staggers easily with a few powerful blows, giving you an opening to land some heavy attacks.
