Fortnite Chapter 5 Season 4 added several new quests for players to complete, including a secret quest that won't initially appear in the menu. Gwenpool's Mission is a found quest, the hidden story quest that you must first unlock by visiting a specific location on the map. Here's how.
Help Gwenpool complete her mission at The Raft Guide – Fortnite Secret Quests
The new update has added The Raft POI to the map, which you’ll need to go to to activate the Gwenpool secret quest. Start a Battle Royale match and glide to the location marked on the map above. There, you’ll find a small, demolished building outside of the stronghold (see second image).
There you will find a door with the Listen prompt as you can see in the image above. Go to the door and interact with it to talk to Gweenpool who happens to be locked in a prison cell on purpose. Continue the conversation and eventually accept the offer to activate the quest you found called Gwenpool's Mission. The quest asks you to help Gwenpool complete her mission at The Raft and has the following simple objectives:
